PROBLEM TO BE SOLVED: To provide compositions and methods for inhibiting Cripto/GRP78 complex formation and signaling.SOLUTION: The invention provides methods and compositions for treating hyperproliferative diseases comprising disrupting Cripto/GRP78 complex formation in a hyperproliferative cell. In certain embodiments, use of an antibody and/or siRNA, optionally coupled with other cancer therapies, can inhibit Cripto/GRP78 binding. Also provided is a method for identifying therapeutic compounds that can selectively inhibit Cripto/GRP78 binding.
