PROBLEM TO BE SOLVED: To provide a method for introducing one or more molecules of interest into a plant cell protoplast.SOLUTION: The method of the invention for introducing a molecule of interest comprises the steps of: preparing a plant cell protoplast; carrying out a first transfection of the plant cell protoplast with a composition capable of altering the regulation of one or more pathways selected from the mismatch repairing system and non homologous end rejoining and/or a composition capable of introducing DSB; carrying out a second transfection to the plant cell protoplast by using one or more molecules of interest such as mutagenic oligonucleotides; and permitting cell wall formation.
