The present invention relates to novel bacterial and mycobacterial compositions containing RNA and cell walls, and methods for making and using these compositions. These compositions have immune stimulating and anti-cancer activity.

1. A method of making a composition comprising isolated mycobacterial RNA associated with mycobacterial cell wall fragments, comprising the sequential steps of:
A) disrupting a mycobacterial cell biomass to generate mycobacterial cell wall fragments, mycobacterial cell wall fragments and associated RNA, disrupted mycobacterial cells, intact mycobacterial cells, and mycobacterial RNA; B) optionally separating intact mycobacterial cells from the generated mycobacterial cell wall fragments and associated RNA; and, C) separating the soluble, cytosolic contents of a portion of the disrupted mycobacterial cells from the generated mycobacterial cell wall fragments and associated mycobacterial RNA to obtain a composition comprising mycobacterial cell wall fragments and associated RNA, wherein the composition comprising the isolated mycobacterial RNA associated with the mycobacterial cell wall fragments does not contain phenol or Streptomyces griseus protease.
