The present invention relates to a method for measuring protein kinase activity, comprising steps of: a) adhering GMBS(N-[γ-maleimidobutyryloxy]sulfosuccinimide ester) on top of a base plate; b) manufacturing a kit for measuring protein kinase activity by adhering a substrate, which reacts to protein kinase on the base plate to which the GMBS has been adhered c) introducing, to the kit, a sample to be analyzed and a mixture of a buffer containing Triton X-100; and d) measuring the activity of the protein kinase by probing a phosphorylation of the substrate caused by protein kinase included in the sample.
