Provided herein are human Toll-like receptor (TLR)-inhibitors and methods for use in individuals having an autoimmune disease or an inflammatory disorder. The TLR inhibitors of the present disclosure are polynucleotides comprising an inhibitory motif for one or more of TLR7, TLR8 and TLR9.

A polynucleotide for use in inhibiting a TLR8-dependent immune response in an individual, wherein the polynucleotide consists of a nucleotide sequence of the formula: 5′-NxX1X2X3X4X5X6-3′, wherein each of N, X1, X2, X3 and X4, is a nucleotide or nucleotide analog, X5 is G or I, X6 is I or A, x is an integer from 0 to 50, and X6 is at the 3′ end of the polynucleotide, with the proviso that when X5X6 is GI or GA, then each of X3 and X4 is A, T or C.
