摘要 The present invention relates to a method for accurately detecting target nucleic acid by asymmetrically and isothermally amplifying target nucleic acid and simultaneously amplifying a signal of a probe by using an external primer set, an internal primer set in which ratios of forward and reverse DNA-RNA-DNA hybrid primers are different, and a DNA-RNA-DNA hybrid signal probe. According to the present invention, compared to a symmetrical iTPA method which is a conventional isothermal primer and probe amplifying method for making the same primer ratio, a signal of a probe can be effectively amplified, and the method of the present invention can be usefully used in studying molecular biology and diagnosing diseases by being applied to accurate detection and confirmation of pathogens, detection of a change in a gene causing a regulated phenotype, diagnosis of receptivity for hereditary disorders, evaluation of gene expression, and various genome projects.
