<p>PROBLEM TO BE SOLVED: To provide a high yield eubacterial minicell producing strain having genetic modification comprising a regulated genetic suicidal mechanism that irreparably destroys parent cell chromosome which enables mini cell production and functional elimination of viable parent cells in a culture at any time during the course of minicell production and purification run.SOLUTION: The invention provides a minicell producing bacterial strain comprising: an expressible gene encoding a minicell producing gene product that modulates one or more of septum formation, binary fission and chromosome segregation; and an expressible gene encoding an endonuclease, where the chromosome of the minicell producing bacterial strain comprises one or more recognition sites of the endonuclease.</p> |
