PROBLEM TO BE SOLVED: To provide a method for use in the detection of a target nucleic acid.SOLUTION: A method comprises the steps of: (i) contacting a single-stranded probe nucleic acid with a sample of interest under conditions effective to generate a probe/target nucleic acid duplex by specific hybridization of the single-stranded probe nucleic acid to a target nucleic acid, if the target nucleic acid is present, wherein a label is attached to the probe/target nucleic acid duplex to result in a detectable change in Raman spectrum of the label by release of the label from the probe/target nucleic acid; (ii) contacting the probe/target nucleic acid duplex with an exonuclease to effect digestion of the duplex and release of the label molecule from the duplex; and (iii) detecting the detectable change in Raman signal of the label at the time of release of the label from the duplex to detect the target nucleic acid, if present. |
