PROBLEM TO BE SOLVED: To provide improved RSV F protein compositions and methods for preparing RSV F protein compositions.SOLUTION: The invention relates to an immunogenic composition comprising one or more RSV F polypeptides, to certain engineered RSV F proteins, and nucleic acids encoding the engineered RSV F proteins. In an embodiment, the RSV F protein is soluble. For example, the transmembrane region and cytoplasmic tail of the RSV F protein may be deleted. In some embodiments, the soluble RSV F has at least one of 1) one or more mutations to one or both furin-cleavage sites; 2) one or more mutations to the fusion peptide; 3) one or more mutations to the p27 linker; 4) an added oligomerization sequence; and 5) an added amino acid sequence that provides a protease cleavage site.
