The present invention relates to a mutant strain with arginine or glutamine production comprising inactivated malic enzyme or lactate dehydrogenase, and to a manufacturing method thereof. The present invention manufactures the mutant strain which does not lower growth than a strain with arginine or glutamine production comprising wild type or activated malic enzyme or lactate dehydrogenase, and has improved arginine or glutamine production and yield per unit time by inactivating malic acid or lactate dehydrogenase, thereby producing arginine or glutamine. Also, productivity and yield per unit time of arginine or glutamine can be improved by using the mutant strain so that costs can be reduced when arginine or glutamine is used as a raw material.
