PROBLEM TO BE SOLVED: To provide a TCR cloning system capable of not only analyzing an unbiased TCR repertoire but also recovering an antigen-specific TCRα/&bgr;cDNA pair, and also evaluating their functions.SOLUTION: There is provided a manufacturing method of a TCR gene specific to antigen a, including: 1) a process of stimulating a T-cell group containing a T-cell specific to the antigen a by an antigen-presenting cell; 2) a process of specifying a T-cell specific to the antigen a from the T-cell group containing the T-cell specific to the antigen a and sorting into a container one by one; and 3) a process of supplying a single activated T-cell specific to the antigen a in the container into a PCR and amplifying the TCR gene specific to the antigen a. According to the present invention, a target TCR gene can be cloned in a shorter period than conventional methods. |
