PROBLEM TO BE SOLVED: To provide compositions of asymmetric interfering RNA and use thereof.SOLUTION: The invention provides asymmetric double-stranded RNA molecules capable of carrying out sequence-specific gene silencing. The RNA molecules comprise the first strand and the second strand. The first strand is longer than the second strand. The RNA molecules comprise double-stranded region formed by the first strand and the second strand, and two termini independently selected from the group consisting of 5'-protruding, 3'-protruding and blunt ends. The RNA molecules of the invention can be used as research tools and/or therapeutic drugs.
