<p>Provided are a detection method and detection kit for detecting a nucleotide mutation simply, with highly sensitivity and high specificity, which allow a plurality of samples to be simultaneously detected in a single measurement. The detection method includes the steps of amplifying a nucleic acid to be tested to obtain a DNA amplification product; subjecting the DNA amplification product, a first nucleic acid probe having a region complementary to a first region of the DNA amplification product, a second nucleic acid probe having a region complementary to a second region of the DNA amplification product, and a plurality of kinds of signal amplification probes to be used in a PALSAR method to a reaction to form a complex for detection including a probe polymer; and detecting the probe polymer bound in the complex for detection to detect the presence or absence of a mutation in a nucleotide mutation site of the DNA amplification product, the first region of the DNA amplification product including the nucleotide mutation site, the nucleotide mutation site being located at a portion other than both end portions of the first region, the second nucleic acid probe having a base sequence identical to part or all of the base sequence of one of the signal amplification probes.</p> |
