A method of mass spectrometry comprises determining a time T (8-11) for calibrating a mass spectrometer during a single experimental run or acquisition, wherein the step of determining comprises determining a time when the mass spectral data being acquired during the experimental run or acquisition is equal to or below a threshold value 7. The mass spectrometer is then calibrated by introducing calibrant or lockmass ions at the time T. The mass spectral data threshold may be applied to total ion current TIC or ion intensities. The invention allows fixed periodic calibration intervals (3-6) to be adjusted so that the calibration times T are chosen to avoid times during which analyte ions of interest (1, 2) are present. |
