The present invention relates to a medium composition for cryopreservation of a human amniotic fluid-derived stem cell. The medium composition is manufactured by the steps of: preparing a cryopreservation solution; deep-freezing AFSCs in the cryopreservation solution obtained from the step above and adding zVAD-fmk in the medium after being thawed; and comparing and assessing a cell survival rate and a cell proliferation rate of AFSCs obtained from the step above and an expression level of mRNA of a stem cell marker. The present invention is capable of reducing cytotoxicity that occurs during freezing and thawing, decreasing physical and chemical damage applied to cells and maintaining a high survival rate of cells.
