Disclosed is Boc (tert-butyloxycarbonyl) protected linagliptin (i.e. 1-[(4-methyl-quinazolin-2- yl)methyl]-3-methyl-7-(2-butyn-1-yl)-8-[(R)-3-(tert-butyloxycarbonylamino)-piperidin-1-yl]-xanthine). Also disclosed is the preparation of a pharmaceutical composition comprising linagliptin, wherein said linagliptin is prepared by a process in which Boc-protected linagliptin is deprotected such as by treatment with trifluoroacetic acid, hydrochloric acid, or by treatment with bromotrimethylsilane or iodotrimethylsilane.
