PROBLEM TO BE SOLVED: To provide an optical observation device, an optical observation method, and an image processing program of a sample observation image, which allow a satisfactory observation image to be obtained by reducing an influence of background light included in an observation image of a sample when the observation image is acquired from the incidence surface side of light to the sample. ! SOLUTION: In a detection system 100, two images (an observation image and a comparison image) are captured by beams emitted to a surface of a cellular structure S at different incidence angles. In respect to the captured image (in a state where target light and background light exist together), an unknown separation value α is defined by input, so that target light and background light included in one image can be separated even when α is unknown.
