A method for expanding epithelial stem cells comprising contacting the stem cells with a culture medium comprising a ErbB3/4 lignad (e.g. neuregulin, heregulin), a receptor tyrosine kinase ligand (e.g. an ErbB3/4 ligand, TGF, FGF, BDNF, PDGF) and a bone morphogenetic protein (BMP) inhibitor (e.g. noggin, chordin, chordin-like protein, follistatin, follistatin-related protein, DAN, sclerostin, decorin, alpha-2 macroglobulin). Also claimed is a culture medium comprising an ErbB3/4 ligand, a receptor tyrosine kinase ligand and a bone morphogenetic protein (BMP) inhibitor. The culture medium optionally comprises, a TGF-β inhibitor, a Notch inhibitor, prostaglandin pathway activator, cAMP pathway activator, BMP pathway activator, a p38 inhibitor, gastrin, nicotinamide, ROCK inhibitor or testosterone. Also claimed is the creation and medical use of organoids, either cancer organoids or lung organoids. Further claimed is a culture medium comprising a p53 stabilising agent. |
