PHOTOACTIVE METHIONINE MIMETICS INTRODUCED PROTEIN G MUTANTS

The present invention relates to protein G mutants with photoactive methionine (pM) mimetics introduced thereto. The protein G mutants are prepared by introducing, into Escherichia coli, the sequence of the third antibody coupled region (PG-C3) of protein G (PG-C3) in which the 32, 35 and 40 positions of PG-C3 are substituted with Met residues and the 37-position are substituted with Arg residues, and plasmids which encode methyonyl tRNA synthase (MRS) mutants. The protein G mutants with pM mimetics introduced thereinto form covalent bonding with antibodies when irradiated with UV rays, so that the protein pM introduced G mutants can be employed usefully in preparing high sensitive biochips and biosensors.
