Provided herein are methods and compositions relating to the synthesis of isoprenoid diphosphates using a mutated isopentenyl phosphate kinase.

1. An isolated protein comprising an amino acid sequence that is at least 90% identical to at least 200 contiguous amino acids of SEQ ID NO: 1;
wherein the amino acid sequence has a substitution at the position corresponding to Ala63 of SEQ ID NO: 1; and wherein said amino acid sequence is capable of catalyzing a reaction between an isoprenoid monophosphate and a phosphate donor to produce an isoprenoid diphosphate.
