The present invention relates to a method for identifying the irradiated and non-irradiated status of pearl nucleus and pearl nacre of a cultured pearl, and more specifically, to a method for identifying the irradiated and non-irradiated status of pearl nucleus and pearl nacre of a cultured pearl, comprising: a step of preparing test samples by grinding cultured pearl nucleus or cultured pearl nacre; a step for dehydrating the test samples; a step of measuring the dried test samples by an electron spin resonance (ESR) meter or an electron paramagnetic resonance (EPR) meter; and a step of checking a g-value gained from measurement and determining the irradiated and non-irradiated status. Establishment or more reliable determination criteria are possible through identification of irradiated and non-irradiated status according to the present invention.
