ANTI-INFALMMATORY PHARMACEUTICAL COMPOSITION CONTAINING CELL-TRANSDUCIBLE PARAOXONASE 1 FUSION PROTEIN

Paraoxonase 1 (PON1) is an antioxidant enzyme which plays an important role in a variety of diseases. However, the mechanism and function of the PON1 are not well-known. The present inventors have prepared a cell-permeable PEP-1-PON1 fusion protein and tested whether the fusion protein shows a protection effect on inflammation in a Raw 264.7 cell treated with LPS or hydrogen peroxide and an animal model with skin inflammation treated with TPA. As a result, the PEP-1-PON1 fusion protein has been effectively transmitted into the RAW 264.6 cell and exhibited a significant cell protective effect on the inflammation and cell death induced by LPS or hydrogen peroxide. In addition, the topically applied PEP-1-PON1 fusion protein has induced a remarkable improvement effect on ear edema induced by 12-O-tetradecanoylphorbol-13-acetate (TPA) and revealed its possible use as a therapeutic agent for a variety of inflammation and oxidative stress-related illnesses.
