PROBLEM TO BE SOLVED: To provide a novel gene marker of Cryptococcus sp. strain S-2, an efficient gene disruption method, and a method for increasing frequency of homologous recombination utilizing such a disruption method.SOLUTION: Provided is a method for disrupting a target gene in the genome of a uracil-requiring strain of Cryptococcus sp. The method comprises a process of preparing a gene construct. The construct comprises a first portion of the 5' region sequence of the target gene, a uracil-auxotrophy marker gene sequence and a first portion of the 3' region sequence of the target gene in this order, and a second portion of the 5' region or of the 3' region of the target gene is inserted between the first portion of the 5' region sequence of the target gene and the uracil-auxotrophy marker gene sequence, and/or between the uracil-auxotrophy marker gene sequence and the first portion of the 3' region sequence of the target gene. |
