Calibrating an apparatus for optically characterising samples of particles of small size comprises using a sample of a calibration population of particles which is substantially monodisperse, homogeneous and has a known concentration of particles per unit volume. The sample is analysed to determine, for a particular combination of apparatus conditions or settings, the number of particles detected and the average brightness of individually detected and measured particles. A new particular combination of apparatus conditions or settings is applied and the analysis is repeated for the same sample, or another from the same calibration population. The analysis may be further repeated for further new combinations of apparatus conditions or settings. A calibration plot or look-up table of brightness of particle against number of particles detected is derived from the analyses, which is used to calibrate the apparatus against subsequent analysis of populations of particles of unknown concentration. The apparatus may be for nanoparticle tracking analysis (NTA). |
