Disclosed herein are anti-TLR3 antibodies as well as methods of making and using them. The antibodies are particularly adapted to the treatment of autoimmune or inflammatory diseases using anti-TLR3 antibodies.

A monoclonal antibody that inhibits TLR3-mediated signaling in a TLR3-expressing cell, wherein the antibody binds to the glycan-free lateral surface of the N-terminal portion of the TLR3 polypeptide, wherein said antibody has reduced binding to a mutant TLR3 polypeptide comprising a mutation at residue 64, 65, 86, 89, 112, 113, 115, 117, 120, 137 and/or 139 of the TLR3 polypeptide of SEQ ID NO: 1, relative to binding between the antibody and a wild-type TLR3 polypeptide of SEQ ID NO: 1.
