The present invention relates to a recellularized bio scaffold and a preparation method thereof. According to the present invention, the recellularized bio scaffold maintains functions of liver cells in a scaffold, has anticancer drug resistance than a two dimensional model, and can effectively copy the surrounding environment of cancer cells, thereby being used for anticancer screening. The preparation method comprises the steps of: flowing a surfactant to the liver of an animal and decellularizing liver; and flowing liver cells to the decellularized liver and recellularizing liver.
