The present invention relates to a method of screening a composition for prevention and treatment of obesity or diabetes, which has determined that higher expression of Htr2a and Htr2b in all tissues of visceral fat, subcutaneous fat, and brown fat in a group of mice which took high-fat diet than in a control group; that the expression of Tph1 also occurred in tissues of a fat cell; that the size of a fat cell in fat tissues of Ap2-Tph1 knock-out mice is diminished; and that injecting p-chlorophenylalanine (PCPA) to obesity-induced mice can achieve reduction in the weight of the mouse, reduction in fat accumulation, inhibition of glucose tolerance defect, reduction in insulin resistance, activation of brown fat, activation of mitochondria, and increase in the metabolic rate, so that the present invention can be employed usefully in screening a composition for prevention and treatment of obesity or diabetes. The method according to the present invention might comprise the steps of: applying a tested substance to a separated fat cell or fat tissue; measuring the expression of Trypotophan hydroxylase (Tph1) from the separated fat cell or fat tissue; and screening the test substance by which the expression of Tph1 is reduced compared to a control group to which the tested substance is not applied. |
