The present invention relates to a preparation method of an organic fertilizer using a byproduct of plant tissue culture fermented by mixing wild ginseng cultural filtrate and a mixed bacterial strain in a mushroom spent medium, and an organic fertilizer by using the byproduct of plant tissue culture prepared by the preparation method. According to the present invention, the organic fertilizer using the ginseng cultural filtrate and the mushroom spent medium byproduct can supply sufficient nutrients to crops when used for a crop cultivation as replaced by agricultural chemicals and fertilizers such that an agricultural product having excellent marketability can be produced and a crop cultivation land can be improved, and also can be used for an eco-friendly organic material agriculture since environmental pollution and ecosystem destruction are prevented by protecting the crops from blight and harmful insects.
