<P>PROBLEM TO BE SOLVED: To provide a means for simply producing a transformant wherein only an objective sequence is inserted into a target site or a target domain on a chromosome. <P>SOLUTION: The method for producing a double-stranded nucleic acid having activity for inserting a first objective nucleotide sequence, a selection marker and a second objective nucleotide sequence into a target site or target domain on the chromosome by a homologous recombination includes: separately synthesizing a sense chain and an anti-sense chain with two kinds of primers, while using a cyclic DNA containing the selection marker and the objective nucleotide sequences as a template, mixing the sense chain with the anti-sense chain, and amplifying the double-stranded nucleic acid.
