FIELD: medicine.SUBSTANCE: invention relates to biotechnology, molecular biology and genetics and represents a method of genetic typing of polymorphism rs2551715 of human glutathione reductase (GSR). The method consists in the genetic typing of polymorphism by a method of a polymerase chain reaction and a method of polymorphism of lengths of restriction fragments with the application of specially selected pair of primers F: 5'-GGAAGAAGCAATGAAATTTGATC-3' and R: 5'-CCGAAACATTTGTGAATCTCC-3' and endonuclease BssT1I for the differentiation of alleles C and T. After amplification with the said pair of primers, PCR product with the size of 138 bp is subjected to hydrolysis. Hydrolysis products are fractioned in a 3% agarous gel, coloured by ethidium bromide and visualised in a transilluminator in passing UV-light. DNA fragments with the size of 114 and 24 bp are verified as the homozygous genotype of a wild type CC, a fragment with the size of 138 bp - as the homozygous mutant genotype TT, and fragments of 138, 114 and 24 bp - as the heterozygous genotype CT.EFFECT: invention makes it possible to simplify the method of the genetic typing of gene GSR rs2551715 polymorphism.1 dwg |
