Disclosed herein are meningococcal immunogenic conjugates which can elicit immune responses against meningococcal polysaccharides (PS) from groups A, C, W-135, and Y and group B factor H binding protein (fHbp). The disclosed conjugates also exhibit bactericidal activity against meningococcal A, C, W-135, Y, B, and X serogroups. Also disclosed are improved methods for preparing conjugates, such as immunogenic conjugates, including activation of a polysaccharide with a cyanylation agent at about 4° C. |

1. A method for preparing an immunogenic conjugate, comprising:
reacting at least one polysaccharide with a cyanylation agent at about 2° C. to about 6° C., resulting in a cyanate-activated polysaccharide; and contacting the at least one cyanate-activated polysaccharide with at least one protein at a pH of about 5 to about 8 at about 2° C. to about 6° C., such that the at least one cyanate-activated polysaccharide reacts with the at least one protein, resulting in an immunogenic conjugate that includes at least one C—N bond formed between the at least one polysaccharide and the at least one protein. |
