Impure solutions of kallikrein inactivator are purified by treating the impure solution with an aqueous solution of a water-soluble salt of metaphosphoric acid whereby the sparingly soluble complex of the inactivator and salt is precipitated, an excess of the metaphosphate salt being avoided, then separating the precipitated complex and freeing it from the salt to give purified solution. Preferred soluble salts are metaphosphates of ammonium or alkali metals, and by avoidance of excess is meant that the aqueous salt solution is added only until no further precipitation occurs. The complex may be freed from the salt by dissolution in a solution containing more than 1% w/v concentration of a salt such as ammonium or sodium chloride or ammonium or alkali metal hydroxide, to decompose it, followed by removal of salts by e.g. dialysis or ion exchange; the pure solution may be lyophilized. Impure solutions for starting materials are suitable preparations from kallikrein-inactivator-containing animal organs, e.g. paratid or lymph glands, lungs, liver, spleen, or pancreas. |
