PROBLEM TO BE SOLVED: To provide a simple and efficient method in which the species of animal hair used for an animal hair product is determined based on its gene information.SOLUTION: There is provided a determining method for animal hair product comprising: 1) a step of cutting or crushing an objective animal hair product; 2) a step of supplying the crushed sample with DNA extracting solution as buffer solution comprising reducing agent and heating it; 3) a step of degrading protein with cysteine protease; 4) a step of extracting DNA from the animal hair product, which comprises steps of denaturing, centrifuging and ultra-filtering supernatant of the reaction solution after protein degradation so that DNA is isolated; 5) a step of carrying out PCR reaction with the isolated DNA as a template, using a set of positive control primer capable of amplifying a common DNA fragment in a plurality of animal species, and a set of primers for determination capable of amplifying a DNA fragment specific for an objective animal species; and 6) a step of confirming the amplified DNA fragment with electrophoresis. In the method, there are provided primers used for determining a yak, cashmere and sheep. |
