PROBLEM TO BE SOLVED: To provide methods for selection of proteases or mutant proteases with desired substrate specificities and activities.SOLUTION: Methods for identifying modified proteases with modified functions comprises screening candidate and modified proteases by contacting them with a substrate, such as a serpin, an alpha macroglobulin or a p35 family protein or a modified serpin and a modified p35 family member or a modified alpha macroglobulin, that, upon cleavage of the substrate, traps the protease by forming a stable complex.
