PROBLEM TO BE SOLVED: To provide an automated device (integrated cartridge) and method for extraction of nucleic acids obtained from cells, amplification of nucleic acids, and detection of nucleic acids.SOLUTION: A device comprises: a housing; an entry port 413 for accepting a sample suspected of containing a target nucleic acid; first chambers 408, 409 containing a reagent for extracting the target nucleic acid; and a first conduit permitting passage of extracted nucleic acid. The housing contains an amplification reagent that is capable of incorporating a detectable label into an amplified nucleic acid target. An amplification chamber 410 has means for controlling amplification conditions, and includes means for moving the amplified target to a sensing region to permit binding to the capture oligonucleotide. The second conduit is attached to a holding chamber which contains a fluid capable of substantially displacing an uncaptured amplified target from the sensing region and permits sensing of the detectable label retained in the sensing region. |
