A process for producing a milk clotting enzyme by fermentation comprises culturing a micro-organism belonging to the genus Irpex, Fomitopsis, Coriolus or Lenzites in a nutrient medium under aerobic conditions, accumulating the milk clotting enzyme in the resultant culture medium, and recovering said enzyme therefrom. The enzyme may be recovered from the culture liquor by precipitation with an organic solvent, salting out, concentration under reduced presture, or absorption and desorption. The crude enzyme so obtained may be dissolved in distilled water and dialysed against flowing water; the purified solution then being freeze-dried.
