Provided are antibody drug conjugates (ADCs), and methods of using such ADCs to treat proliferative disorders. Specifically, the disclosure provides engineered antibody immunospecifically reacts with a determinant of DLL3, SEZ6 or CD324. The antibodies comprising one or more unpaired cysteine residues, which provide site(s) for the selective and controlled conjugation to a therapeutic or diagnostic agent.
