Glycine transporter-1 inhibitors, methods of making them, and uses thereof

Disclosed are sulphonyl substituted heteroalkylcyclic or alkylcyclic benzamides of formula (I) where the substituents are as disclosed in the specification. Examples of compounds of formula (I) are: 2-Amino-6-chloro-N-((1-(4-(propylsulfonyl)piperazin-1-yl)cyclohexyl)methyl)benzamide; 2,4-Dichloro-N-((1-(4-(propylsulfonyl)piperazin-1-yl)cyclohexyl)methyl)benzamide; N-((1-(4-(Propylsulfonyl)piperazin-1-yl)cyclohexyl)methyl)-2-(trifluoromethoxy)benzamide; 3-Chloro-2-fluoro-N-((1-(4-(propylsulfonyl)piperazin-1-yl)cyclohexyl)methyl)-6- (trifluoromethyl)benzamide and N-((1-(4-(Propylsulfonyl)piperazin-1-yl)cyclohexyl)methyl)-4-(trifluoromethyl)benzamide. Also disclosed are compositions comprising compounds of formula (I). Further disclosed is the use of the compounds in the manufacture of a medicament for inhibiting GlyT-1.
