#CMT# #/CMT# Examining a human sample for markers of liver diseases comprises detecting elevated levels of one or more proteins selected from ER60, vimentin, actin alpha 1 skeletal muscle protein, hMFAP 4, tropomyosin, PTGES 2, amyloid P components, transgelin, calponin 1, p20, 17 kD myosin light chain, H chain H Igg B12 and prolyl 4-hydroxylase beta subunit, or detecting decreased levels of one or more proteins selected from methylene tetrahydrofolate dehydrogenase 1, PRO2619, aldehyde dehydrogenase 1, fibrinogen alpha chain prepro protein, fructose bisphosphate aldolase B, argininosuccinate synthetase, Eef1a2, ATP5A1, alpha-2 actin, regucalcin, serum albumin, mitochondrial malate dehydrogenase and acetoacetyl-CoA thiolase. #CMT#USE : #/CMT# Diagnosing liver diseases, especially hepatic fibrosis. #CMT#INSTRUMENTATION AND TESTING : #/CMT# Preferred Method: The proteins are detected by bioanalytic methods, e.g. immunohistochemistry, antibody arrays, Luminex, enzyme-linked immunosorbent assay (ELISA), immunofluorescence or radioimmunoassay, or by mass-spectrometric methods, e.g. multi reaction monitoring or absolute quantification, or by 2D electrophoresis with isoelectric focussing in the first dimension and gel electrophoresis in the second dimension. The samples are labeled with a dye, especially Cy2, Cy3 or Cy5, before electrophoresis. The sample is a liver biopsy sample or a whole blood, serum or plasma sample. #CMT#EXAMPLE : #/CMT# The ER60 protein level in fibrotic liver cells is 26.9 times higher than in normal cells.
