PROBLEM TO BE SOLVED: To provide novel Stat3 pathway inhibitors, and to provide cancer stem cell inhibitors.SOLUTION: The present invention relates to: a novel naphtho class of compounds as Stat3 pathway inhibitors and as cancer stem cell inhibitors; methods of using such compounds to treat cancer; methods of using such compounds to treat disorders in a mammal related to abnormal Stat3 pathway activity; and pharmaceutical compositions containing such compounds. In one embodiment, the present invention provides a method of treating disorders related to an abnormal Stat3 pathway activity in a subject, comprising administering to the subject an effective amount of a compound of formulae I-VIII as described in the specification.
