<p>A method of depleting non desired nucleic acid sequences from a nucleic acid library comprising generating a library of single stranded DNA fragments with fixed orientation adapters attached to each end, annealing a probe specific for the non desired sequences and wherein at least one of the end adaptors has a restriction endonuclease recognition sequence, extending the probe with a DNA polymerase to generate a double stranded DNA fragment that extends to one of the adapters, restriction digesting the double stranded nucleic acid at the adaptor at the restriction endonuclease sequence and performing PCR with primers specific to the adaptors , wherein only undigested DNA sequences are amplified. Also claimed are kits and methods for performing the invention, which may be performed on sorted cell or single cell populations.</p> |
