Purification is by passing a soln. of lysozyme contaminated with protein impurities through a column packed with a polyacrylamide/agarose gel in pearl form, and collecting the eluate only after a vol. of soln. equal to the vol. of the pearls has passed through the column. Protein rapidly passes through the gel which acts as a molecular sieve. Lysozyme is retained for some time and then issues freed from protein. The prefd. gel pearls are of 100-160 mu m in dia. and contain 6% acrylamide and 4% agarose. Lysozyme is sepd. from egg albumin readily using this method.
|
