<p>1311726 13-Aminomethyl-steroids SCHERING AG 17 April 1970 [17 April 199] 18520/70 Heading C2U Novel steroids of the formula and salts thereof, wherein R 1 is =O, H(#-OZ 1 ) or H(α-OZ 1 ) in which OZ 1 is a free, esterified or etherified OH group, OR 2 is a free, esterified or etherified OH group, R 3 is H or alkoxy, R 4 is H or CH 3 , R 5 is -CH 2 CH 2 OZ 2 in which OZ 2 is a free, esterified or etherified OH group, or -NR 4 R 5 is 2-oxo-oxazolidin-1-yl, C 4 =C 5 is a single bond when R 1 is OH or a derivative thereof and a single or double bond when R 1 is =O, and C 5 =C 6 is a single or double bond when R 1 is OH or a derivative thereof or a single bond when R 1 is =O, any H atom at position -5 havingα- or #-configuration, are prepared by splitting by reduction a steroid of the formula when required treating the product of the formula with phosgene or a carbonic acid ester to give, after possible saponification in the 3- and/or 20-position, a steroid of the formula in which R<SP>1</SP> 1 and R<SP>1</SP> 2 are each H or -CO-O alkyl, esterifying a free 20-OH group, and when required reducing with a complex metal hydride to give a steroid of the formula and when required esterifying or etherifying a free OH group in these products and/or converting by oxidation a 3-ol to a 3-one or a #<SP>5</SP>-3-ol to a #<SP>4</SP>-3-one. Salts are prepared by standard procedures. Steroids of the second general formula above are prepared by known methods from 3-acetoxy- 16α-R 3 -pregnan-20-ols or -#<SP>5</SP>-pregnen-20-ols. Thus 3#-acetoxy-5#-pregnan-20#-ol with I 2 and CaCO 3 in cyclohexane under reflux and with irradiation, with subsequent heating with acetic acid and sodium acetate, gives 3#- acetoxy - 18,20# - epoxy - 5# - pregnan - 18 - ol and this with ethanolamine gives 18-(2<SP>1</SP>- hydroxyethylamino) -18,20#-epoxy-5#-pregnan- 3# - ol. 16α- methoxy - 18 - (2<SP>1 </SP>- hydroxyethylamino) - 18,20α- epoxy - 5#-pregnan - 3#- ol is prepared similarly via 16α-methoxy-3#- acetoxy - 18,20α- epoxy - 5# - pregnan - 18 - ol, the starting 16α-methoxy-3#-acetoxy-5#-pregnan- 20α-ol being prepared from 3#-acetoxy-5#- pregn - 16 - en - 20 - one via 16α- methoxy- 3#-acetoxy-5#-pregnan-20-one. The steroids of the invention are stated to have favourable activity on the cardiac muscle tonus during the diastole, and they may be made up into pharmaceutical compositions with suitable carriers.</p> |
