Grafts, systems, methods and kits are provided for treating a subject having a defect using adult stem cells that are characterized in that the cells do not substantially induce a T cell or natural kill (NK) cell-mediated immune response in the subject. The systems, methods and kits optionally further include exogenous biological factors and energy sources such as an ultrasound device for stimulating healing of the target tissue and tissue defect and/or tissue formation.
