PROBLEM TO BE SOLVED: To provide antibodies against MUC16 and use methods thereof.SOLUTION: The invention provides an antibody and an antigen-binding fragment thereof that specifically bind a polypeptide or its antigenic portion, where the polypeptide is selected from a) MUC16 ectodomain polypeptide, b) MUC16 cytoplasmic domain polypeptide and c) MUC16 extracellular domain polypeptide containing a cysteine loop polypeptide. The invention's antibody and composition containing it are useful for diagnostic and therapeutic applications for diseases in which MUC16 is overexpressed, such as cancer.
