Disclosed is an isolated monoclonal antibody or an antigen-binding fragment thereof, which specifically binds to the extracellular domain of EP4 expressed on the cell surface of mammalian cells, and inhibits the cellular function of EP4 to increase the intracellular cAMP level and does not bind to EP1, EP2 or EP3. Also disclosed is the use of the monoclonal antibody or an antigen-binding fragment thereof for the treatment of EP4-related immunological diseases, a tumor or pain.
