An autoantibody detection method for detecting, with exceptional accuracy, an autoantibody causing an autoimmune disease is provided. An autoantibody detection method according to the present invention is characterized by the inclusion of a contact step for causing an antigen reagent including a denatured protein presented by a major histocompatibility complex (MHC) class II molecule and a sample to come into contact with each other and a detection step for detecting a complex of an autoantibody in the sample and the denatured protein in the antigen reagent. Further, using this detection method to detect a complex of the autoantibody and the denatured protein in a biological sample isolated from a subject makes it possible to test for the possibility of the contraction of an autoimmune disease by the subject on the basis of the detection results.
