The present invention relates to a method for extracting and purifying at least one target nucleic acid contained in a specimen, comprising the following steps: (a) placing the specimen in contact with a lysis buffer optionally containing glycerol, as a result of which a cellular lysate is obtained; (b) placing the cellular lysate obtained after step (a) in contact with magnetisable particles, under conditions making it possible for said particles to capture said at least one target nucleic acid; (c) washing said magnetisable particles obtained after step (b) with a washing buffer containing glycerol; (d) placing said washed particles obtained after step (c) in contact with an elution buffer, under conditions allowing the release of said target nucleic acid in the elution buffer. The present invention also relates to said lysis and washing buffers. |
