A method of providing a human variable heavy chain acceptor framework for the grafting of lagomorph heavy chain CDRs, comprising changing the amino acid at one or more of positions 24, 56, 84, 89 and 108 (AHo numbering) of a human variable heavy chain framework sequence so that the resulting framework sequence comprises at least four of the following amino acids: threonine (T) at position 24, alanine (A) or glycine (G) at position 56, threonine (T) at position 84, valine (V) at position 89, and arginine (R) at position 108 (AHo numbering), whereby the resulting human variable heavy chain acceptor framework is an acceptor for the heavy chain CDRs from a donor lagomorph immunobinder. |
