PROBLEM TO BE SOLVED: To provide methods of using sequence-based analysis and rational strategies to modify and improve the structural and biophysical properties of immunobinders, in particular of single chain antibodies (scFvs), including such properties as stability, solubility, and/or antigen binding affinity, and to provide immunobinders produced by the engineering methods.SOLUTION: An immuno binder, particularly an scFv is identified by amino acid residues preferred for substitution through analysis of a database of stable scFv sequences selected, and engineered by performing one or more substitutions at identified amino acid positions. The scFv antibody is produced using preferred scFv framework scaffolds which can insert a CDR sequence and such preferred framework scaffolds. |
